Un hombre ha desaparecido sin dejar rastro en Nueva York. Klute (Donald Sutherland), un policia íntimo amigo suyo, encuentra una pista en unas cartas que aquel le escribía a una prostituta llamada Bree (Jane Fonda), así que va a buscarla para interrogarla, iniciando con ella una relación amorosa tempestuosa. Simultaneamente, Klute prosigue con sus investigacioens en los bajos fondos cuando, de pronto, las prostitutas empiezan a ser asesinadas, y Klute piensa que Bree puede ser la próxima víctima.
